Europska agencija za lijekove (EMA) jučer je dala zeleno svjetlo za odobrenje novog cjepiva protiv COVID-a - proizvođača Novavax. Nakon što mu Europska komisija da službeno odobrenje, Nuvaxovid će biti peto cjepivo protiv koronavirusa koje će se moći koristiti u zemljama Europske unije. I prije odobrenja EU je potpisala ugovor o kupnji 200 milijuna doza ovog cjepiva.

Riječ je o potpuno novom cjepivu koje nije vektorsko (kao što su AstraZeneca i Janssen) niti je rađeno najnovijom mRNA tehnologijom ( Pfizer i Moderna), nego o tzv. proteinskom cjepivu. Radi se o prokušanoj metodi koja je testirana na brojnim drugim cjepivima, prisutnima na tržištu desecima godina. Ako je skepticima i ono problem, onda su im i problem sva druga cjepiva koja su već testirana godina na milijunima ljudi. Novavax je koristio sličnu tehnologiju kao cjepiva za hepatitis B i hripavac, koja se ne moraju skladištiti na izrazito niskim temperaturama. 

Ovo cjepivo sadrži laboratorijski uzgojenu verziju proteina šiljka koji je prisutan na površini virusa SARS-CoV-2. Cjepivo također sadrži "adjuvans", tvar koja pomaže pojačati imunološki odgovor na cjepivo. Kada se cjepivo primijeni, imunološki sustav osobe identificirat će protein kao strani te će protiv njega stvoriti prirodnu obranu, odnosno antitijela i T stanice. Ako kasnije cijepljena osoba dođe u kontakt s virusom SARS-CoV-2, imunološki će sustav prepoznati protein šiljka na virusu i biti spreman za napad. Protutijela i imunološke stanice zajednički djeluju na ubijanju virusa, sprječavanju njegovog ulaska u stanice tijela i uništavanju zaraženih stanica, čime pomažu u zaštiti od COVID-19.

Primjenjuje se u dvije doze uz vremenski razmak od tri tjedna između doza.

Rezultati kliničkih podataka iz dvaju glavnih kliničkih ispitivanja pokazali su da je Nuvaxovid siguran i djelotvoran u prevenciji korone u osoba u dobi od 18 i više godina. Ova ispitivanja obuhvatila su više od 45.000 ispitanika. Dvije trećine ispitanika koji su sudjelovali u prvom ispitivanju primilo je cjepivo, a jedna trećina placebo. Polovina ispitanika obuhvaćena drugim ispitivanjem primila je cjepivo, a druga polovina placebo.

Prvo kliničko ispitivanje, provedeno u Meksiku i Sjedinjenim Američkim Državama, pokazalo je smanjenje broja simptomatskih slučajeva zaraze u osoba koje su primile cjepivo Nuvaxovid od 90,4 posto sedam dana nakon primjene druge doze.

Kod osoba koje su cijepljene - njih 14 od 17.312 dobilo je simptome COVID-a, a kod ispitanika koji su primili samo placebo, 63 od ukupno 8.140 ispitaniih  je dobilo simptome korone.

Drugo kliničko ispitivanje, provedeno u Ujedinjenom Kraljevstvu, također je pokazalo slično smanjenje broja simptomatskih slučajeva zaraze u osoba koje su primile Nuvaxovid. Samo od 10 od 7.020 ispitanika dobilo je simptome COVID-a, dok je kod 7019 onih koji su primili placebo njih 96 oboljelo. U tom ispitivanju cjepivo je pokazalo djelotvornost od 89,7 posto.

Ono što je manje ohrabrujuće u ovim istraživanjima djelotvornosti cjepiva jest činjenica da se ona rađena na izvornoj varijanti virusa, te na soju alfa i beta, a kako djeluje na danas dominantnu deltu i omikron - ne znamo.

- Trenutačno su dostupni ograničeni podaci o djelotvornosti cjepiva Nuvaxovid protiv drugih varijanti koronavirusa, uključujući varijantu omikron - tumače iz HALMED-a.

U kliničkim testiranjima cijepljenih nuspojave kod cijepljenih su bile blage, od boli i osjetljivost na mjestu primjene cjepiva, umoru, bolovima u mišićima, glavobolji, općoj slabosti, bolovima u zglobovima te mučnini. Prolazili su za najkasnije dva dana.

Iz HALMED-a navode kako će se sigurnost i djelotvornost cjepiva nastaviti intenzivno pratiti i nakon dolaska na tržište EU-a, posebice kroz sustav prikupljanja i analize prijava sumnji na nuspojave te putem dodatnih studija proizvođača i europskih nadležnih tijela.

Europska agencija za lijekove (EMA) jučer je dala zeleno svjetlo za odobrenje novog cjepiva protiv COVID-a - proizvođača Novavax. Nakon što mu Europska komisija da službeno odobrenje, Nuvaxovid će biti peto cjepivo protiv koronavirusa koje će se moći koristiti u zemljama Europske unije. I prije odobrenja EU je potpisala ugovor o kupnji 200 milijuna doza ovog cjepiva.

Riječ je o potpuno novom cjepivu koje nije vektorsko (kao što su AstraZeneca i Janssen) niti je rađeno najnovijom mRNA tehnologijom ( Pfizer i Moderna), nego o tzv. proteinskom cjepivu. Radi se o prokušanoj metodi koja je testirana na brojnim drugim cjepivima, prisutnima na tržištu desecima godina. Ako je skepticima i ono problem, onda su im i problem sva druga cjepiva koja su već testirana godina na milijunima ljudi. Novavax je koristio sličnu tehnologiju kao cjepiva za hepatitis B i hripavac, koja se ne moraju skladištiti na izrazito niskim temperaturama. 

Ovo cjepivo sadrži laboratorijski uzgojenu verziju proteina šiljka koji je prisutan na površini virusa SARS-CoV-2. Cjepivo također sadrži "adjuvans", tvar koja pomaže pojačati imunološki odgovor na cjepivo. Kada se cjepivo primijeni, imunološki sustav osobe identificirat će protein kao strani te će protiv njega stvoriti prirodnu obranu, odnosno antitijela i T stanice. Ako kasnije cijepljena osoba dođe u kontakt s virusom SARS-CoV-2, imunološki će sustav prepoznati protein šiljka na virusu i biti spreman za napad. Protutijela i imunološke stanice zajednički djeluju na ubijanju virusa, sprječavanju njegovog ulaska u stanice tijela i uništavanju zaraženih stanica, čime pomažu u zaštiti od COVID-19.

Primjenjuje se u dvije doze uz vremenski razmak od tri tjedna između doza.

Rezultati kliničkih podataka iz dvaju glavnih kliničkih ispitivanja pokazali su da je Nuvaxovid siguran i djelotvoran u prevenciji korone u osoba u dobi od 18 i više godina. Ova ispitivanja obuhvatila su više od 45.000 ispitanika. Dvije trećine ispitanika koji su sudjelovali u prvom ispitivanju primilo je cjepivo, a jedna trećina placebo. Polovina ispitanika obuhvaćena drugim ispitivanjem primila je cjepivo, a druga polovina placebo.

Prvo kliničko ispitivanje, provedeno u Meksiku i Sjedinjenim Američkim Državama, pokazalo je smanjenje broja simptomatskih slučajeva zaraze u osoba koje su primile cjepivo Nuvaxovid od 90,4 posto sedam dana nakon primjene druge doze.

Kod osoba koje su cijepljene - njih 14 od 17.312 dobilo je simptome COVID-a, a kod ispitanika koji su primili samo placebo, 63 od ukupno 8.140 ispitaniih  je dobilo simptome korone.

Drugo kliničko ispitivanje, provedeno u Ujedinjenom Kraljevstvu, također je pokazalo slično smanjenje broja simptomatskih slučajeva zaraze u osoba koje su primile Nuvaxovid. Samo od 10 od 7.020 ispitanika dobilo je simptome COVID-a, dok je kod 7019 onih koji su primili placebo njih 96 oboljelo. U tom ispitivanju cjepivo je pokazalo djelotvornost od 89,7 posto.

Ono što je manje ohrabrujuće u ovim istraživanjima djelotvornosti cjepiva jest činjenica da se ona rađena na izvornoj varijanti virusa, te na soju alfa i beta, a kako djeluje na danas dominantnu deltu i omikron - ne znamo.

- Trenutačno su dostupni ograničeni podaci o djelotvornosti cjepiva Nuvaxovid protiv drugih varijanti koronavirusa, uključujući varijantu omikron - tumače iz HALMED-a.

U kliničkim testiranjima cijepljenih nuspojave kod cijepljenih su bile blage, od boli i osjetljivost na mjestu primjene cjepiva, umoru, bolovima u mišićima, glavobolji, općoj slabosti, bolovima u zglobovima te mučnini. Prolazili su za najkasnije dva dana.

Iz HALMED-a navode kako će se sigurnost i djelotvornost cjepiva nastaviti intenzivno pratiti i nakon dolaska na tržište EU-a, posebice kroz sustav prikupljanja i analize prijava sumnji na nuspojave te putem dodatnih studija proizvođača i europskih nadležnih tijela.
